Improvement in group identification of dojo loach, Misgurnus anguillicaudatus, using PCR-restriction fragment length polymorphism

چکیده

In most Japanese populations of dojo loach (Misgurnus anguillicaudatus), gonochoristic diploids genetically diversified groups (A and B, further subdivided into B1 B2) are present, whereas unisexual clonal lineages inhabit certain localities in the Hokkaido Ishikawa Prefectures Japan. Through a series genetic studies including DNA markers, loaches were deemed to originate from hybridization event(s) between A groups. However, combined analyses with other markers needed identify each group. this study, we improved PCR-restriction fragment length polymorphism (RFLP) analysis recombination activating gene 1 (RAG1) using digestion two restriction enzymes, PvuII StuI. The RAG1-RFLP showed different patterns for group: fragments (245 198 bp) group A, three (198, 147, 98 B1, single (443 B2. exhibited four (245, 198, derived both B1. Moreover, able detect hybrid genotypes × B2 B2). Thus, allowed quick accurate identification loaches.

Developmental transcriptome analysis and identification of genes involved in formation of intestinal air-breathing function of Dojo loach, Misgurnus anguillicaudatus

Dojo loach, Misgurnus anguillicaudatus is a freshwater fish species of the loach family Cobitidae, using its posterior intestine as an accessory air-breathing organ. Little is known about the molecular regulatory mechanisms in the formation of intestinal air-breathing function of M. anguillicaudatus. Here high-throughput sequencing of mRNAs was performed from six developmental stages of posteri...

Identification of infectious agents in onychomycoses by PCR-terminal restriction fragment length polymorphism.

A fast and reliable assay for the identification of dermatophyte fungi and nondermatophyte fungi (NDF) in onychomycosis is essential, since NDF are especially difficult to cure using standard treatment. Diagnosis is usually based on both direct microscopic examination of nail scrapings and macroscopic and microscopic identification of the infectious fungus in culture assays. In the last decade,...

Identification of nontuberculous mycobacteria existing in tap water by PCR-restriction fragment length polymorphism.

This paper presents the finding of the possible cause of the high false-positive rate in acid-fast staining in histological examinations. Using acid-fast staining, culture, and PCR, acid-fast bacilli were detected in 83.7% of 49 hospital tap water samples and nontuberculous mycobacteria (NTM) were detected in 20.4% of the same 49 samples. The 10 NTM isolates were also identified to the species ...

Restriction Fragment Length Polymorphism (RFLP)

Restriction Fragment Length Polymorphism (RFLP) is a technique in which organisms may be differentiated by analysis of patterns derived from cleavage of their DNA. If two organisms differ in the distance between sites of cleavage of a particular restriction endonuclease, the length of the fragments produced will differ when the DNA is digested with a restriction enzyme. The similarity of the pa...

Identification of Staphylococcus spp. by PCR-restriction fragment length polymorphism analysis of dnaJ gene.

A PCR-restriction fragment length polymorphism (RFLP) analysis method that analyzes a part of the dnaJ gene was designed for the rapid and accurate identification of Staphylococcus spp. XapI or Bsp143I digestion of the PCR-generated products rendered distinctive RFLP patterns that allowed 41 reference species and subspecies to be identified with a high degree of specificity. The novel method wa...
